| Disease | TAXOL IS AN ANTITUBULIN AGENT WITH A UNIQUE MODE OF ACTION IN THAT IT ENHANCES RATHER THAN INHIBITS TUBULIN POLYMERIZATION. THE EFFECT ON TUBULIN-GTP INTERACTIONS IS HIGHLY DEPENDENT ON REACTION CONDITIONS. TUBULIN POLYMERIZATION USUALLY REQUIRES HYDROLYSIS, BUT TAXOL CAN ELIMINATE THIS GTP REQUIREMENT. UNDER REACTION CONDITIONS IN WHICH TUBULIN WILL NOT ORDINARILY POLYMERIZE,HOWEVER, TAXOL WILL INDUCE POLYMERIZATION ONLY IN THE PRESENCE OF GTP. GTP HYDROLYSIS BY TUBULIN UNDER THESE SAME CONDITIONS IS COMPLETELY INDEPENDENT ON TAXOL. |
