LUBROL-PX TREATMENT OF THE MYOCARDIAL SARCOLEMMA INHIBITED F- AND FORSKOLIN-STIMULATED ADENYLATE CYCLASE ACTIVITIES. SIMILIAR INHIBITION OF F-,FORSKOLIN AND GUANINE NUCLEOTIDE RESPONSIVE ADENYLATE CYCLASE ACTIVITIES WAS ALSO OBSERVED IN LUBROL-PX SOLUBILIZED MEMBRANES. WHEN GUANINE NUCLEOTIDE REGULATORY PROTEIN WAS REMOVED FROM THE 100,000 X G SUPERNATANT FRACTION BY GTP-SEPHAROSE AFFINITY CHROMATOGRAPHY,THE STIMULATION WAS ALMOST COMPLETELY ABOLISHED. PHOSPHOLIPASE A-2 AND C TREATMENTS OF THE MYCOCARDIAL SARCOLEMMA RESULTED IN A SIGNIFICANT DECREASE IN F- AND FORSKOLIN-STIMULATED ADENYLATE CYCLASE ACTIVITIES. THESE DATA SUGGEST THAT GUANINE NUCLEOTIDE REGULATORY PROTEIN AND PHOSPHOLIPIDS MAY BE REQUIRED FOR F- AND FORSKOLIN ACTIVATION OF ADENYLATE CYCLASE IN HEART SARCOLEMMA.
