| Disease | DOSE-INDEPENDENT, TWO COMPARTMENT PLASMA KINETICS WERE OBSERVED. PLASMA CONCENTRATIONS OBTAINED UP TO 10 DAYS SUGGEST A THIRD, TERMINAL ELIMINATION PHASE. WITHIN 10 DAYS, 47% OF THE DOSE WAS EXCRETED IN THE URINE AND FECES. THREE HOUR BILIARY EXCRETION STUDIES ACCOUNTED FOR 1.7% OF THE DOSE. LIVER, SKELETAL MUSCLE, KIDNEY AND FAT OBTAINED 24 HOURS AFTER DOSING CONTAINED 19, 14, 6 AND 4% OF THE DOSE, RESPECTIVELY. CARDIAC MUSCLE AND BRAIN EACH CONTAINED LESS THAN 1% OF THE ADMINISTERED DOSE. THE RAPID DISTRIBUTION INTO TISSUES AND SLOW ADMINISTRATION SUGGEST THAT MULTIPLE DOSES OF EMETINE MAY LEAD TO ACCUMULATION AND CHRONIC TOXICITY. |
