DiseaseGOSSYPOL-ACETIC ACID WAS USED IN THE STUDY CELLULAR DAMAGE IN GERMINAL EPITHELIUM OF TESTES WAS OBSERVABLE FOLLOWING 2-3 WKS OF GOSSYPOL ADMINISTRATION.BY THE END OF 4-5 WKS SPERMATIDS + SPERMATOCYTES OF MID + LATE STAGES HAD ALMOST DISAPPEARED COMPLETELY. THE SEVERLY DAMAGED SEMINIFEROUS TUBULES BECAME ATROPHIC AND DEPOPULATED TO A MARKED DEGREE,AND ONLY A SINGLE LAYER OF CELLS CONSISTING OF SERTOLI CELLS AND SPERMATOGONIA WERE LEFT IN THE TUBULES. THERE WAS NO DETECTABLE DAMAGE IN BOTH THE LEYDIG INTERSTITIAL CELLS AND THE EPIDIDYMAL EPITHELIA IN GOSSYPOL-TREATED RATS.MOREOVER,NO CHANGES IN ACCESSORY GLANDS WERE OBSERVED. CYTOLOGICAL + AUTORADIOGRAPHIC STUDIES ON INCORPORATION OF 14-C LABELLED AMINO ACIDS INDICATED THAT SPERMATIDS + SPERMATOCYTES OF MID + LATE STAGES WERE MOST SENSITIVE AND VULNERABLE OF THE SPERMATOGENIC SERIES IN RESPONSE TO GOSSYPOL THEY WERE FIRST TO SHOW DETECTABLE DAMAGE AND FIRST TO DEGERERATE.ELECTRON MICROSCOPY DEMONSTRATED THE ULTRASTRUCTURAL CHANGES IN THESE TWO TYPES OF GERM CELLS.THE MOST OBVIOUS DAMAGE WAS IN THE MITOCHONDRIA OF SPERMATIDS SPERMATOGONIA MAINTAINED ABILITY TO INCORPORATE 3-H THYMIDINE THOUGH SEVERE DAMAGE OCCURRED IN SEMINIFEROUS TUBULES,INDICATING THAT THERE WAS APPRECIABLE DIFFERENCE IN SENSITIVITY BETWEEN SPERMATOGENIC CELLS IN RESPONSE TO GOSSYPOL THE EFFECT,SITE,AND THE POSSIBLE PATHWAY OF THE ANTISPERMATOGENIC ACTION OF GOSSYPOL,TOGETHER WITH ITS SIGNIFICANCE TO CONTRACEPTION WERE ANALYZED AND DISCUSSED.
