| Disease | MATURE ANIMALS WERE DOSED WITH 5.0 MG EACH DAILY FIVE TIMES EACH WEEK FOR LONG TERMS.RATS WERE SACRIFICED AFTER1,2,3,4 AND 6 MONTHS DOSING AND WITHDRAWAL FOR 1,2 AND 3 MONTHS.THE TESTES WERE EXAMINED HISTOLOGICALLY. RATS DOSED MORE THAN 2 MONTHS DEVELOPED TESTICULAR ATROPHY,BUT NOT IN ALL ANIMALS. THE NUMBER AND DEGREE OF DEGENERATION OF TESTIS WERE CLOSELY CORRELATED TO THE LENGTH OF DURATION OF TREATMENT. THIS SEEMED TO INDICATE THAT TESTIS ATROPHY MIGHT BE CAUSED BY GOSSYPOL DIRECTLY.HOWEVER,THE OCCURRENCE OF DAMAGED SPERMATIDS AT STEPS 18 AND 19 WERE REALLY EARLIER THAN TESTICULAR ATROPHY. THE FIRST SIGN OT THE LATTER WAS THE ACCUMULATION OF DEAD SPERMS IN THE SAMINIFEROUS TUBULE ADJACENT TO RETE TESTIS AND DEGRENERATION ALWAYS BEGAN FROM THERE. IT MAY BE POSTULATED THAT TESTICULAR ATROPHY IS CAUSED BY THE DEAD SPERM ACCUMULATED IN THE TUBULES. |
