DiseaseDAILY DOSING FOR 7 WEEKS PRODUCED STRIKING ULTRASTRUCTURAL DEFECTS AS EARLY AS 3 WEEKS AFTER THE START OF DOSING. BY THE 5TH WEEK SIGNIFICANT DAMAGE TO VIRTUALLY ALL SPERM FLAGELLA WAS OBSERVED THROUGHOUT THE EPIDIDYMAL DUCT OF ALL TREATED ANIMALS. THE INTIAL AND PREDOMINANT DEFECT WAS DEGENERATION OF THE MIDPIECE MITOCHONDRIA. CAUDAL SPERMATOZOA WERE TOTALLY IMMOBILE BY THE 5TH WEEK. THE ULTRASTRUCTURE OF THE EPIDIDYMAL AND VASAL EPITHELIUM WAS NOT AFFECTED BY GOSSYPOL TREATMENT. IN THE TESTIS THE LIGHT MICROSCOPIC EFFECT OF GOSSYPOL TREATMENT COULD BE DETECTED IN ONLY SMALL PROPORTION OF THE SEMINIFEROUS TUBULES. HISTOLOGICAL CHANGES INCLUDED INTER-AND INTRACELLULAR VACUOLIZATION, PRIMARILY IN THE SERTOLI CELLS,LEADING TO GIANT CELL FORMATION. EXFOLIATION AND TUBULAR ATROPHY AT LATEER TIME INTERVALS. AT NO TIME DURING THE STUDIES WERE THESE CHANGES UNIFORMLY DISTRIBUTED THROUGHOUT THE TESTIS. WITH ELECTRON MICRSCOPE, ULTRASTRUCTURAL ANALYSIS REVEALED SPECIFIC DEGENERATIVE CHANGES IN THE MITOCHONDRIAL SHEATH OF LATE SPERMATIDS. OTHER COMPONENTS OF THE FLAGELLUM DID NOT APPEAR TO BE AFFECTED. NO OTHER DIFFERENCES BETWEEN GERM CELLS OF CONTROLS AND GOSSYPOL-TREATED ANIMALS COULD BE DETECTED. THE ULTRASTRUCTURE OF THE LEYDIG CELLS AND OTHER INTERSTITAL ELEMENTS APPEARED NORMAL.
