| Disease | THIS PAPER REPORTS THE RESULTS OF OBSERVATION BY MEANS OF HISTOCHEMICAL,ELECTRON MICROSCOPIC,CYTOCHEMICAL AND BIOCHEMICAL TECHNIQUES DESIGNED TO INVESTIGATE THE EFFECT OF GOSSYPOL ON NA-K-ATPASE ACTIVITY IN RENAL TUBULES AND GASTROCNEMIUS MUSCLE. IT WAS SHOWN THAT INTENSIVE CYTOCHEMICAL REACTION OF NA-K-ATPASE ACTIVITY WAS OBSERVED IN GASTROCNEMIUS SARCOLEMMA,BASAL + INTERDIGITATING MEMBRANES OF DISTAL CONVOLUTED TUBULE, + CELL MEMBRANES OF THE THICK ASCENDING SEGMENT OF THE KIDNEY. NA-K-ATPASE ACTIVITY OF RENAL TUBULES + GASTROCNEMIUS MUSCLE WAS MARKEDLY DECREASED AT THIS DOSE LEVEL. OBVIOUS INHIBITION IN NA-K-ATPASE OF RENAL CELL MEMBRANE FRACTION ISOLATED FROM TREATED ANIMALS FED ON THE SAME DOSAGE WAS ALSO DEMONSTRATED. IT IS SUGGESTED THAT GOSSYPOL MAY INDUCE THE REDUCTION OF NA-K-ATPASE ACTIVITY AND THE DEGREE OF REDUCTION IS DOSE DEPENDENT. THE CYTOCHEMISTRY OF THE MEMBRANE NA-K-ATPASE AND THE MECHANISM OF GOSSYPOL INHIBITION OF ACTIVITY WAS DISCUSSED. |
