| Disease | IN CLINICAL TRIALS THE SPERM COUNT DECREASED AND ABNORMAL SPERM WERE DETECTED IN SEMEN AFTER 4 WEEKS OF GOSSYPOL ADMINISTRATION. BY THE 8TH WEEK OF ADMINISTRATION, THE SPERM COUNT WAS MARKEDLY DECREASED WHEREAS EXFOLIATED SPERMATOGENIC CELLS (INCLUDING MAINLY SPERMATIDS AND SPERMATOCYTES) INCREASED CONCOMMITANTLY. THE SITE OF GOSSYPOL ACTION WAS AT THE SPERMIOGENIC SPERMATIDS AS WELL AS THE MID AND LATE-STAGE SPERMATOCYTES. GOSSYPOL DECREASED THE ACTIVITY OF SUCCINATE DEHYDROGENASE IN THE MIDDLE PIECE OF THE SPERM AND DAMAGED THE SPIRAL MITOCHONDRIAL SHEATH. THE EFFECT ON ACROSOMAL PROTEINASE APPEARED TO BE SECONDARY. THE ULTRASTRUCTURAL CHANGES OF SPERM NUCLEAR CHROMATIN AFTER GOSSYPOL TREATMENT SUGGESTED THAT GOSSYPOL MAY AFFECT THE METABOLISM OF NUCLEIC ACIDS. |
