| Disease | DAILY DOSING FOR 7 WEEKS. SOURCE OF TEST COMPOUND WAS THE PEOPLE'S REPUBLIC OF CHINA. FOUND TO BE GREATER THAN 98% PURE BY HPLC AT NICHD. ULTRASTRUCTURAL DEFECTS WERE FOUND IN EPIDIDYMAL SPERM, BUT NOT IN EPIDIDYMAL EPITHELIUM, IN THE INITIAL SEGMENT OF THE CAPUT EPIDIDYMIS AS EARLY AS 3 WEEKS AFTER START OF DOSING. MINOR DEGENERATIVE CHANGES WERE OBSERVED IN A FEW SPERMATOZOA FOR AS LITTLE AS 2 WKS OF TREATMENT. THE SEVERITY AND FREQUENCY OF THESE CHANGES APPEAR TO INCREASE WITH THE DOSE AND DURATION OF TREATMENT. BY THE FIFTH WEEK SIGNIFICANT DAMAGE TO VIRTUALLY ALL SPERM FLAGELLA WAS OBSERVED THROUGHOUT THE EPIDIDYMAL DUCT OF ALL TREATED ANIMALS. CAUDAL SPERMATOZOA EXAMINED AT VARIOUS TIME INTERVALS WERE TOTALLY IMMOBILE. |
