| Disease | DAILY DOSING FOR 28 DAYS PRODUCED A RISE IN THE SODIUM CONCENTRATION BUT A FALL IN SPERM CONCENTRATION IN THE CAUDAL EPIDIDYMAL FLUIDS. THE EPIDIDYMAL SPERMATOZOA ALSO LOST THEIR CAPACITY TO INTIATE MOTILITY UPON SUSPENSION IN A SODIUM CONTAINING MEDIUM. TO SEE WHETHER THESE CHANGES WERE ATTRIBUTABLE IN PART TO A DIRECT EFFECT OF GOSSYPOL ON THE EPIDIDYMAL EPITHELIUM AND/OR EPIDIDYMAL SPERM,THE EPIDIDYMAL CONTENT AND SPERMATOZOA WERE ISOLATED FROM THE TESTIS BY LIGATION. THE OPERATION ALONE DID NOT AFFECT NA+,K+ & SPERM CONCENTRATIONS OF CAUDA EPIDIDYMAL FLUID,EXCEPT THAT SPERM CONCENTRATION OF 17 AND 34 DAY LIGATED EPIDIDYMIS WAS SIGNIFICANTLY LOWER THAN THAT OF UNLIGATED EPIDIDYMIS. DOSING DAILY FOR UP TO 34 DAYS DID NOT RESULT IN A CHANGE IN THE SODIUM,POTASSIUM AND SPERM CONCENTRATIONS OF THE EPIDIDYMAL FLUID AND MOTILITY INITIATION OF SPERMATOZOA WHICH HAD BEEN RETAINED IN THE CAUDA EPIDIDYMIDES FOR UP TO 34 DAYS. ANALYSIS OF PROTEINS BY SDS-GEL ELECTROPHORESIS HAS SHOWN THAT THE PROTEIN PATTERNS OF FLUIDS FLUSHED OUT FROM THE CAUDA EPIDIDYMIDES OF RATS WERE QUALITATIVELY SIMILAR TO THOSE OF THE NORMAL RATS. IT WAS CONCLUDED THAT GOSSYPOL ACETIC ACID HAS NO DIRECT EFFECT ON TRANSPORT PROCESSES OF CAUDA EPIDIDYMIDES AND CHANGES IN THE EPIDIDYMAL NA+ CONCENTRATION & SPERM FUNCTION COULD BE ATTRIBUTABLE TO AN EFFECT AT THE MORE PROXIMAL SITES. |
