DiseaseDAILY DOSING FOR SEVEN WEEKS. THE STUDY CONFIRMED AND EXTENDED EARILER OBSERVATIONS DEMONSTRATING THAT EPIDIDYMAL SPERM OF TREATED RATS EXHIBITED DISTINCT ULTRASTRUCTURAL CHANGES. THE SEVERITY AND FREQUENCY OF THE DEGENERATIVE CHANGES APPEAR TO INCREASE WITH THE DOSE AND DURATION OF TREATMENT. STRIKING ULTRASTRUCTURAL DEFECTS CAN BE SEEN AS EARLY AS 3 WEEKS AFTER THE START OF DOSING AT 20 MG/KG. BY THE FIFTH WEEK OF EITHER 20 OR 30 MG/KG OF GOSSYPOL, SIGNIFICANT DAMAGE TO VIRTUALLY ALL SPERM FLAGELLA IS OBSERVED THROUGHOUT THE EPIDIDYMAL DUCT OF ALL TREATED RATS. THE INITIAL AND PREDOMINANT DEFECT IS DEGENERATION OF THE MIDPIECE MITOCHONDRIA BUT ADDITIONAL FLAGELLARR DEFECTS OCCUR. CAUDAL SPERMATOZOA ARE TOTALLY IMMOTILE BY THE FIFTH WEEK. THE ULTRASTRUCTURE OF THE EPIDIDYMAL AND VASAL ELITHELIUM IS NOT AFFECTED BY GOSSYPOL. IN THE TESTIS, THE LIGHT MICROSCOPIC EFFECTS OF GOSSYPOL TREATMENT CAN BE DETECTED IN ONLY A SMALL PROPORTION OF THE SEMINIFEROUS TUBULES. HISTOLOGICAL CHANGES INCLUDE INTER- AND INTRACELLULAR VACUOLIZATION, PRIMARILY IN THE SERTOLI CELLS. THIS LEADS TO GIANT CELL FORMATION, EXFOLIATION AAND TUBULAR ATROPHY AT LATER TIME INTERVALS. HOWEVER, AT NO TIME ARE THESE CHANGES UNIFORMLY DISTRIBUTED THROUGHOUT THE TESTIS. ULTRASTRUCTURAL ANALYSIS REVEALS SPECIFIC DEGENERATIVE CHANGES IN THE MITOCHONDRIAL SHEATH OF LATE SPERMATIDS. OTHER COMPONENTS OF THE FLAGELLUM DO NOT APPEAR TO BE AFFECTED. NO OTHER DIFFERENCES BETWEEN GERM CELLS OF CONTROL AND GOSSYPOL-TREATED RATS HAVE BEEN DETECTED. THE ULTRASTRUCTURE OF THE LEYDIG CELLS AND OTHER INTERSTITIAL ELEMENTS ALSO APPEARS NORMAL.
