DiseaseTESTES FROM ANIMALS DOSED DAILY FOR 1-12 WEEKS WERE STUDIED IN THIN SECTIONS AND FREEZE FRACTION REPLICAS. THE ANTIBIOTIC FILIPIN WAS USED AS A TRACER TO TEST THE PERMEABILITY OF THE BARRIER. SOME LEYDIG CELLS AND 10% OF THE SERTOLI CELLS DEVELOPED LARGE AUTOPHAGIC VACUOLES AND INCREASED CYTOPLASMIC ELECTRODENSITY,WHILE OTHER SERTOLI CELLS SHOWED AN ANTI-LH EFFECT VIA CLUMPS OF HETEROCHROMATIN LINING THE NUCLEUS. MANY SERTOLI-SERTOLI CELL JUNCTIONS WERE VACUOLIZED AND DISTENTED: INTERRUPTED JUNCTIONAL STRANDS SERVED AS A NIDUS FOR MULTIPLE "ATYPICAL RECTANGULAR GAP JUNCTIONS". IN REGIONS OF INTACT JUNCTIONS,FILIPIN WAS ACCESSIBLE TO THE BASAL STRUCTRUES,BUT IT WAS EXCLUDED FROM THE INTERMEDIATE COMPARTMENT FORMED BY THE LUMINAL AND BASAL SERTOLI-JUNCTIONAL STRANDS. IN MEMBRANE DOMAINS CONTAINING DISRUPTED JUNCTIONS,FILIPIN PERTURBATIONS WERE OBSERVED BOTH WITHIN AND AROUND THE INTERMEDIATE COMPARTMENT. THESE FOCAL PERMEABILITY ALTERATIONS ARE INTERPRETED AS A LOSS OF COMPARTMENTAL ORGANIZTION ALONG THE SEMINIFEROUS TUBULES. GOSSYPOL DISRUPTION OF SERTOLI CELLS AND SERTOLI-SERTOLI CELL JUNCTIONAL INTEGRITY RASES DOUBTS ABOUT THE REVERSIBILITY OF THIS EXPERIMENTAL ANTI-FERTILITY COMPOUND.
