| Disease | ETOPOSIDE LEVELS WERE DETERMINED IN 6 PATIENTS WITH GESTOCHORIOCARCINOMA UNDERGOING SURGERY. A SINGLE 100 OR 200 MG/M2 DOSE WAS GIVEN BY ONE HOUR INFUSION PRIOR TO SURGERY. THE TERMINAL PLASMA HALF LIFE WAS 4.1 HOURS, VOLUME OF DISTRIBUTION WAS 9 L/M2, AND CLEARANCE WAS 21.5 ML/MIN./M2. CONCENTRATION IN MYOMETRIAL CARCINOMA AND NORMAL MYOMETRIUM WERE 40-50% AT 50 AND 740 MINUTES AFTER INFUSION. AFTER 25-180 MINUTES THE CONCENTRATION IN LUNG METASTASIS WAS 19-43%, AND IN NORMAL LUNG 38-61% OF THE PLASMA LEVELS. LEVELS IN SUBCUTANEOUS TISSUE WERE ONLY 5-27% OF PLASMA LEVEL. IN 24 HOUR URINE 19-44% OF THE DRUG WAS UNCHANGED, AND 5-22% WAS CONJUGATED WITH GLUCURONIC ACID OR SULFATE. |
