| Disease | COMBINATION CHEMOTHERAPY WITH CISPLATIN WAS USED AS SALVAGE THERAPY IN 30 PATIENTS WITH PREVIOUSLY TREATED SMALL CELL LUNG CANCER. TREATMENT CYCLES WERE 28 DAYS AND THE FOLLOWING SCHEDULES WERE USED: IN 8 PATIENTS, 150 MG/SQ.M. OVER 30 MINUTES DAYS 1-3 WITH CISPLATIN 20 MG/SQ.M DAYS 1-5. IN 15 PATIENTS 115 AND 15 MG/SQ.M. OF ETOPOSIDE AND CISPLATIN RESPECTIVELY AND IN 7 PATIENTS 115 AND 25 MG/SQ.M. OF ETOPOSIDE AND CISPLATIN DAYS 1-3 RESPECTIVELY. THERE WERE 2 COMPLETE AND 6 PARTIAL RESPONSES, FOR A MAJOR RESPONSE RATE OF 27%. MINOR RESPONSES WERE SEEN IN 7 OTHER PATIENTS. TOXIC EFFECTS INCLUDED MILD NAUSEA AND VOMITING. THE DOSE LIMITING TOXICITY WAS MYELOSUPPRESSION. LUEKOPENIA WAS COMMON WITH 6 MILD AND 5 SEVERE CASES. THROMBOCYTOPENIA WAS SEEN IN PATIENTS UNDER THE FIRST TREATMENT SCHEDULE ONLY. NEUROTOXICIGY INCLUDED ONE CASE OF IRREVERSIBLE SENSORY MOTOR NEUROPATHY INVOLVING TIBIAL AND PERSONEAL NERVES AFTER 6 CYCLES, AND ONE CASE OF CLINICALLY APPARENT HEARING LOSS. THERE WAS ALSO SOME HEPATOTOXICITY AND MILD ALLERGIC REACTION ATTRIBUTABLE TO CISPLATIN THERAPY. THE AUTHORS RECOMMEND THE THIRD TREATMENT SCHEDULE FOR FURTHER CLINICAL TRIALS. |
