| Disease | PHASE I CLINICAL TRIAL OF COMPOUND IN TREATING CANCER. FIFTY-ONE PATIENTS WITH CANCER NOT AMENABLE TO STANDARD THERAPIES WERE ENROLLED. ESCALATING DOSES FROM 60-2000 MG/M2 WERE BOLUS ADMINISTERED. DYSPNEA EXPERIENCED AT HIGHEST DOSE. AT DOSES ABOVE 945 MG/M2, EMESIS OCCURRED IN 33% OF COURSES AND WAS NOT ALLIEVIATED BY SEROTONIN ANTAGONISTS. 3 OF 14 PATIENTS AT DOSE LEVEL 945 MG/M2 DEVELOPED ACUTE RENAL INSUFFICIENCY, INDEPENDENT OF DOSING INTERVAL (1 WEEK OR 3 WEEKS). THIS EFFECT WAS MODERATED BY PREHYDRATION. NO PATIENTS ACHIEVED RADIOLOGICAL RESPONSE TO TREATMENTS BUT ONE WITH HEPATOCELLULAR CARCINOMA SHOWED DECREASED ALPHA FETO PROTEIN AND ALKALINE PHOSPHATASE AND ONE WITH OVARIAN CARCINOMA SHOWED DECREASED CA-125 LEVELS. |
