DiseaseDURING THE STEADY-STATE STUDY, 6 DIFFERENT RABBITS (3.5-4 KG) WERE GIVEN DAILY SUBCUTANEOUS INJECTIONS OF WARFARIN 0.6 MG/KG/DAY FOR 7 DAYS. ON DAY 4, THE RABBITS WERE GIVEN DONG QUAI 2 G/KG ORALLY 2 X/DAILY FOR 3 DAYS WHILE RECEIVING DAILY SUBCUTANEOUS WARFARIN. DAILY BLOOD SAMPLES WERE OBTAINED AND PROCESSED AS IN THE SINGLE-DOSE STUDY. IN THE SINGLE-DOSE STUDY, DONG QUAI ALONE DID NOT AFFECT PT VALUES DURING THE 3-DAY TREATMENT PERIOD. NO SIGNIFICANT DIFFERENCES (P<0.05) WERE FOUND IN DERIVED PHARMACOKINETIC PARAMETERS FOR WARFARIN ALONE AND WITH DONG QUAI. IN THE STEADY-STATE STUDY, MEAN PT VALUES WERE SIGNIFICANTLY INCREASED (P<0.05) AFTER ONLY 2 DAYS OF CONCURRENT DONG QUAI. AFTER 4 DAYS OF CONCURRENT DONG QUAI, THE VALUES TRIPLED (30 SEC) COMPARED WITH BASELINE (10 SEC). NO SIGNIFICANT ALTERATIONS (P<0.05) WERE FOUND IN AVERAGE PLASMA WARFARIN STEADY-STATE CONCENTRATIONS AFTER DONG QUAI ADMINISTRATION. ONE RABBIT DIED ON DAY 6 AND 1 ON DAY 7, BUT CAUSE OF DEATH WERE NOT STATED. FROM THESE RESULTS, IT IS CONCLUDED THAT DONG QUAI COADMINISTERED WITH WARFARIN AT STEADY STATE SHOWED EVIDENCE OF A PHARMACODYNAMIC, NOT A PHARMACOKINETIC INTERACTION.
