| Disease | SILYMARIN IS POORLY ABSORBED FROM THE GI TRACT, WITH BIOAVAILABILITY ESTIMATED AT 23-47%. PEAK PLASMA CONCENTRATIONS OCCUR 2 TO 4 HOURS AFTER AN ORAL DOSE. THE ELIMINATION HALF-LIFE IS APPROXIMATELY 6 HOURS. SILYMARIN APPEARS TO UNDERGO EXTENSIVE ENTEROHEPATIC CIRCULATION, ACHIEVING BILLARY LEVELS SEVERAL TIMES GREATER THAN THOSE IN SERUM. SOM 20-40% OF AN ORAL DOSE IS RECOVERABLE FROM THE BILE AS GLUCURONIDE AND SULFATE CONJUGATES, WITH ONLY 2-5% EXCRETED IN THE URINE. PHOSPHATIDYLCHOLINE SEEMS TO ENHANCE THE ORAL ABSORPTION OF SILYMARIN. IN A STUDY OF 9 CHOLECYSTECTOMY PATIENTS WITH T-DUBE DRANAGEM, BILIARY CONCENTRATIONS AFTER A SINGEL ORAL DOSE OF SILYBIN INCREASED FROM 3% TO 11% WHEN AN EQUIVALENT DOSE OF PHOSPHATIDYLCHOLINE-BOUND SILYBIN WAS USED. |
