FIFTEEN ADULT MALE CHICKENS WEIGHING 900-1200G WERE USED IN THE STUDY AN DWERE RANDOMLY DIVIDED INTO THREE EQUAL GROUPS DESIGNATED 1,2 AND 3. RESULTS SHOWED THAT GRAPEFRUIT JUICE INCREASED PLASMA CONCN. LEVELS BY 7-37%. IN VITRO STUDIES INDICATED CYP3A4 AND CYP2D1/6 AS THE TWO MAJOR ISOFORMS INVOLVED IN CHLOROQUINE METAB. THE INCREASE IN THE CONCN. OF CHLOROQUINE OF PLASMA OF THE CHICKENS THAT RECEIVED GRAPEFRUITS WAS IN LINE WITH THIS. CIMETIDINE IS A KNOWN GENERAL INHIBITOR OF CYTOCHROME P 450 ENZYMES AND IS EXPECTED TO DEPRESS THE METAB. OF CHLOROQUINE BY THE LIVER. THEREBY PROLONGING THE STAY OF THE DRUG IN THE BODY AND RAISING THE PLASMA LEVELS. RELATIVELY SLIGHT INCREASES IN CHLOROQUINE LEVELS WERE HOWEVER, NOTED IN BIRDS PRETREATED WITH CIMETIDINE. THIS SEEMED TO SUGGEST THAT CERTAIN CYTOCHROME P450 ENZYMES WERE NOT INHIBHITED, OR WERE LESS AFFECTED BY THE INHIBITORY ACTION OF CIMETIDINE. |
