DiseaseER-POSITIVE MAMMARY GLAND TUMORS WERE INDUCED IN 5 GROUPS OF 14-15 FEMALE 50-DAY-OLD SPRAGUE-DAWLEY RATS BY A SINGLE ORAL APPLICATION OF 20 MG DMBA. AFTER 5-9 WEEKS NEOPLASTIC GROWTH, THE ANIMALS WERE OVARIECTOMIZED (OVX). BEGINNING AT DAY 10 AFTER OVX, ONE GROUP RECEIVED DAILY DOSES OF 450 GAMMA MESTRANOL/KG BW. 3 GROUPS WERE TREATED P.O. WITH CR IN DOSES COMPARABLE TO 1X, 10X OR 100X HUMAN THERAPEUTIC DOSAGE. THE FIFTH GROUP SERVED AS THE VEHICLE CONTROL. AFTER 7 WEEKS, THE ANIMALS WERE SACRIFICED AND TUMOR NUMBER AND SIZE WERE RECORDED. PLASMA HORMONE LEVELS WERE ANALYZED. CESSATION OF ENDOGENOUS ESTROGEN PRODUCTION AFTER OVX RESULTED IN REGRESSION OF MAMMARY TUMORS IN ALL ANIMALS. ESTROGEN SUBSTITUTION WITH MESTRANOL CAUSED A CLEAR RE-INCREASED IN TUMOR SIZE. AT NECROPSY, NO CLEAR-CUT DIFFERENCE IN TUMOR NUMBER AND SIZE COULD BE DETECTED BETWEEN THE CR GROUPS AND THE VEHICLE CONTROL. NO STIMULATION OF MAMMARY GLAND CANCER RE-GROWTH WAS SEEN IN ANY CR GROUP. IN THE CR GROUP A TREND TOWARDS TUMOR SIZE REDUCTION COMPARED TO THE VEHICLE CONTROL COULD EVEN BE SEEN. NO ESTROGEN AGONISTIC EFFECTS ON PLASMA HORMONE LEVELS (PROLACTIN, FSH, LH) AND ON RELATIVE OR ABSOLUTE ORGAN WEIGHTS COULD BE DETECTED. HISTOLOGY REVEALED NO ESTROGENIC INFLUENCE OF CR ON THE UTERINE TISSUE.
